A comparative study was conducted, considering the hepatoprotective effect of polyprenols (PP) extracted from Siberian fir, and other standard types of treatment for alcoholic hepatitis on the ability to stimulate the processes of natural liver regeneration in rats.
It demonstrated that during administration of PP into rats along with ethyl alcohol the activity of lipid peroxidation processes (LPO) increased to a lesser extent. Having a certain antioxidant effect, PPs prevent the development of negative changes in phospholipid metabolism, inhibiting the formation of lysophospholipid fractions that have a detergent effect on the hepatic parenchyma.
The studied PPs had a preserving effect on hepatocyte metabolism, which manifested itself in the maintenance of glycogen synthetic processes. Also PPs promoted restoration of bile secretion processes in rats with alcoholic liver damage, had a pronounced normalizing effect on the content of bile acids and cholesterol in bile, and the percentage of bilirubin in bile in rats, which received the studied PPs against the background of alcohol exposure, did not differ from the normal during the experiment.
In general, a comparison of the hepatoprotective activity of PP in conditions of alcoholic liver damage in rats showed a comparable focus of their therapeutic effect with the effect of the medicine karsil. These materials offer prospects for the use of PPs as agents of choice in the treatment of alcoholic liver damage, which is highly relevant.
In the study, a patented remedy for stimulation of natural liver regeneration processes, Ropren, was used, which is a PP derived from woody greens of Siberian fir, providing preservation of the organ stroma without significant connective tissue overgrowths, activation of metabolic processes in the absence of activation of spontaneous pathology. In a study conducted on rats, it was demonstrated that when animals are intoxicated with dichloroethane and phenacetin, their body weight significantly decreases, and the relative weight of the liver increases. Treatment of animals with PP allowed to regain body weight and relative liver weight to normal values more frequently than treatment with the drugs legalon and essenciale recommended for therapeutic practice. When treated with this drug and comparison drugs, it was noted that all of them reduced biochemical signs of liver damage, but compared to legalon, the use of PP and essenciale showed a higher therapeutic effect. In general, administration of polyprenols significantly improved morphological, biochemical and functional indices of the liver in experimental animals and prevented their death. Polyprenols in many indicators shows superior or similar results in, reparative processes in comparison with essenciale, and is significantly superior to legalon in terms of the studied indicators.
This is confirmed by histological studies of the liver of intact and experimental animals. Thus, all the morphological features of regenerative changes noted above indicate the development of more effective reparative processes in the liver after the introduction of PP.
